Carbamoylated Erythropoietin-Induced Cerebral Blood Perfusion and Vascular Gene Regulation

Cerebral hypoperfusion is associated with enhanced cognitive decline and increased risk of neuropsychiatric disorders. Erythropoietin (EPO) a neurotrophic factor known to improve function in preclinical clinical studies neurodegenerative psychiatric However, the application EPO limited due its erythropoietic activity that can adversely elevate hematocrit non-anemic populations. Carbamoylated erythropoietin (CEPO), chemically engineered non-erythropoietic derivative EPO, does not alter maintains behavioral effects comparable EPO. Our study aimed investigate role CEPO cerebral hemodynamics. Magnetic resonance imaging (MRI) analysis indicated blood perfusion hippocampal striatal region without altering tight junction integrity. In vitro vivo analyses neurotransmission was unaltered likely EDRF, CGRP, NOS-mediated vasodilation. using human umbilical vein endothelial cells (HUVEC) vascular gene expression showed be non-angiogenic agent which regulates MEOX2 expression. The results from our demonstrate novel modulating vasodilation perfusion.
